Öğe Boğalarda Bovine Herpes Virus Tip 1 (Bhv-1) Enfeksiyonunun Serolojik ve Virolojik Olarak Araştırılması(Selçuk Üniversitesi, 1998) Yavru, Sibel; Şimşek, Atilla; Öztürk, FeridunA lolal of 300 blood sera samples laken from bulls in variaus ages and breed in Meat and Fish Association's Slaughterhouse in Konya were tested against Bovine Herpes Virus type 1 (BHV-') antibodies by mitroneutralisation test. 125 (41.66 %) sera were found to be positive . The neutralising antibody titers ol the posilive sera were detected between 1:2.37 and 1:200. The highest antibody incidance (91.1%-100%) and titers were observed in bulls over 3 years old. In addilion, conjunctival or nasal swap samples from 15 bul1s with respiraıoric symptoms in Slaughterhouse were taken for virus isolation. A virus was isolated by inoculation onto ce1l culture. The isolated virus was identified BHV-' by cross microneutralisation tesl for used hyperimmun sera obtained from rabbits.Öğe Boğalarda bovine herpes virus tip 1 (BHV-1) enfeksiyonunun serolojik ve virolojik olarak araştırılması(1998) Yavru, Sibel; Şimşek, Atilla; Öztürk, FeridunKonya Et ve Balık Kurumu mezbahasına getirilen farklı ırk ve yaşlardaki toplam 300 adet boğadan alınan kan serumu örnekleri Bovine Herpes Virus type 1 (BHV-1 )'e karşı mikronötralizasyon testi ile kontrol edildi. Test sonunda 125 (% 41.66) adet kan serumu seropozitif olarak bulundu. Pozitif serumların nötralizan antikor titrelerinin 1:2.37 ile 1:200 arasında olduğu saptandı. En yüksek antikor dağılımının (% 93.1-%100) ve antikor titrelerinin 3 yaşın üzerindeki boğalarda olduğu tespit edildi. Ayrıca Konya Et ve Balık Kurumu mezbahasına getirilen solunum yolu enfeksiyonuna bağlı klinik belirti gösteren 15 boğadan konjunktival ve nazal swap örnekleri izolasyon materyali olarak alındı. Bu örneklerin hücre kültürlerine inokulasyonu sonucu 1 adet virus izole edildi. İzole edilen virus tavşanlardan hazırlanan hiperimmun serumla çapraz mikronötralizasyon testi sonuçlarına göre BHV-1 olarak identifiye edildi.Öğe Burdur Bölgesi? Süt Sığırcılığı I?şletmeleri?nde Sığır Lökemi Vi?rüs (Blv) Enfeksi?yonunun Prevalansi(Selçuk Üniversitesi, 2004) Kale, Mehmet; Öztürk, FeridunBurdur ili ve ilçelerinde yetiştirilen, klinik açıdan sağlıklı ve ileri gebe olan 469 adet 3 yaş ve üzeri Holstein ırkı süt sığını Bovine Lökemi Virus (BLV) enfeksiyonu yönünden serolojik olarak ELISA-süt ve AGID-kan ile test edildi. Dört yüz altmış dokuz kan serumunun 23'ünde (%4.9), süt serumunun 90 inda (%19.2) BLV'una karşı antikor varlığı tespit edildi. AGID testi ile en az seropozitif hayvan sayısı (1) Burdur-Merkez M3, Burdur-Yeşilova Y1, Y2, Burdur-Tefenni T2, T3 ve en fazla seropozitif hayvan sayısı (11) Burdur-Merkez M2 işletmelerinde belirlendi. Burdur-Merkez M1 ve Burdur-Ağlasun A1 işletmelerinde hiç seropozitif hayvan tespit edilemedi. ELISA testi ile en az seropozitif hayvan sayısı (3) Burdur-Yeşilova Y1 ve en fazla seropozitif hayvan sayısı (17) Burdur-Tefenni T1 işletmelerinde belirlendi. Hem AGID hem de ELISA ile en fazla BLV ile enfekte hayvan sayısının, Burdur-Merkez M2, Burdur-Tefenni T1 ve Burdur-Ağlasun A2 işletmelerinde bulunduğu belirlendi. Enfekte hayvan sayısı yüksek olan bu işletmelerde ortak-karışık emzirme, suni tohumlama öncesinde yapılan sık tedavi ve rektal muayene uygulamalarının yaygın olarak yapıldığı belirlendi.Öğe Kli?ni?k Olarak Sağlıklı Sığır Sürüleri?nde Persiste Bovi?ne Vi?ral Diarrhea Virus Enfeksi?yonlarının Araştırılması ve Epi?zooti?yoloji?k Önemi?(Selçuk Üniversitesi, 1997 Aralık) Şimşek, Atilla; Öztürk, FeridunA total of 142 clinically healthy cattle in five herds were examined serologically and for persistent infection with Bovine Viral Diarrhea Virus (BVDV) in Konya. Leucocyte and blood sera samples were obtained from the ani. mals. BVDV antigens were determined in total of two animals in two herds after examination by immunoflouresans test (IFT) of the 142 leucocyte samples. Second leucocyte samples were obtained from 2 cattle two months after the initial sampling and BVDV antigens were not detected. The 142 first blood sera samples were tested against BVDV by sera neutralisation (SN) test. Antibodies to BVDV were detected in 113 (%79. 5) of 142 samples. Although viremic cattle and high seropositivity were found in two herds, persistent infected (Pl) cattle couldn't be detected in this study.Öğe Konya Bölgesi? The cattle blood sera collected from 950 cattle in Meat and Fish Association's Slaughterhouse in Konya, were subjected to microneutralization test, initially by diluting 1/10, then by diluting them two times and by comparing BAV type 2 viruses with the ratio of 100 DKID 501.05 ml. MDBK (Madine Darby Bovine Kidney) cell culture were used in virus production, in titration and in microneutralization test. As a cell medium, Eagle 's MEM with % 10 inactivated calf sera and as virus medium Eagle's MEM medium without sera were used. At the end of the test, at undiluted blood sera, the 134 cattle blood sera (56.30 %) were found seropositive for the neutralizing antibodies to IBR| IPV virus. The highest incidence of antibody was found in cattle over two years old. The 61 (45.52 %) of the 134 titrated positive sera showed a neutralizing antibody-titers in a range of 1:39.9 and 1:317. With this aim, the 79 cattle blood sera were collected and examined for antibodies to PIV-3 by microneutralisation test. The blood sera collected from 214 cattle were subjected to micro serum neutralization test, initially by diluting 1/10, then by diluting them two times and by comparing BAV type 1, type 2 and type 3 viruses with the ratio of 100 DKID 50/0.05 ml. Based on the results the neutralizing antibodies were found in 153 sera against BAV type 1, in 179 sera against BAV type 2 and in 191 sera against BAV type 3. These findings pointed out that the adenovirus infections which are one of the significant breeding diseases were prevalent in cattle in Konya Agricultural Institution.Öğe Koyunlarda Mavi? The blood sera were examined to determine the presence of neutralizing antibodies to BTV (Bluetongue Virus), type SA4 by microneutralization test. BHK (Baby Hamster Kidney) permanent cell line was used for production and titration of BTV and neutralisation test. Glascow Eagle's MEM (Minimum Essential Medium) with 10% calf sera for cell medium and Glascow Eagle's MEM without sera for virus medium were used in this research. The blood sera were tested at the rate of 1/10 dilution. Bu serumlarda üst solunum yolu enfeksiyonları viruslarından parainfluenza-3 virus (PIV-3), infectious bovine rhinotracheitis virus (IBRV), bovine viral diarrhea-mucosal disease virus (BVD/MDV), bovine adenovirus tip 1,2,3 (BAV-1,2,3), ovine adenovirus tip 1,3 (OAV-1,3), mavi dil virus (BTV) ve bovine respiratory syncytial virus (BRSV) 'larına karşı nötralizan antikorların varlığı mikronötralizasyon testi (MNT) ile araştırıldı. Nontoxic viıamın C (ascorbic acid) concentrations for MDBK were not cause a deereasa in titer of viruses.Öğe Sığır Vebası Şüpheli Buzağılardan Vi?rus I?zolasyonu(Selçuk Üniversitesi, 1993) Öztürk, Feridun; Yavru, Sibel; Şimşek, Atilla; Erer, HüdaverdiA highly lethal disease, suspected of resembling Rinderpest, was diagnosed as a result of clinical and pathological examinations, in calves, in october 1991 in Konya Total of 9 specimens, including 2 mesenterial lymph nodes, 2 spleens, 2 buffy coat samples and 3 nasal swaps was obtained for virology testing from Rinderpest suspected calves The inoculation of specimens to Foetal Calf Kidney (FCK) cell cultures, showed that, 4 of the specimens of the 2 mesenterial lymph nodes and 2 nasal swaps was positive for cytopathologic effect (CPE) The histopathologic examination of specimens with CPE in cell cultures, revealed intrastoplasmic and int- ranuclear inclusion bodies.Öğe Türkiye'de Klinik Olarak Sağlıklı Koyunlardan I?zole Edilen Koyun Adenovirus Tip 3 (OA) Üzerinde Araştırmalar(Selçuk Üniversitesi, 1987 Ocak) Öztürk, FeridunA cytopathogen virus isolated by means of MDBK (Madin Darby Bovine Kidney) and primary lamb kidney tissue cultures from the faeces of clinically healthy sheep brought to Meat and Fish Organization Ankara. Isolated virus was also able to multiply in cattle originated cells besides the sheep originated ones and the type of its nucleic acid was DNA. According to measurements made by electronmicroscope its length was about 100 milimicron. It was resisted against fat soluables such as ether and chloroform and also trypsin enzyme. It lost infectiousness at 56°C in 120 minutes and at 60°C in 30 minutes. In between 3.0 and 4.0 pH degrees its infectiousness estimated to be decreased. It gave heamagglutination with rat erythrocytes and no clinical symptoms estimated in rabbit, quinea pig and mice, but neutralizing antibodies were estimated in rabbit and quinea pig. Neutralisation estimated with OA, by means of cross neutralisation. It is concluded that the isolated virus was ovine adenovirus type 3 (OA) in accordance to the characteristics mentioned above.
