    In this study early pregnancy diagnosis rate was determined in ewes that previously synchronised by MAP, PMSG and GnRH and bred naturally in late anestrous season. Fourty Merino ewes, 5 years old, were used as materials. Jugular blood samples were collected on 15 th, 16 th and 17 th days, after breedings. Samples were santrifuged in 2000 rpm and plasmas were stored at -20°C until assay procedures. For the early pregnancy diagnosis, plasma progesterone and oestradiol 17 B levels were determined by RIA method. For this purpose, Mini assay type Gamma counter and 1251 Progesterone and Oestradiol 17 B assay kits were used. Laboratory results were controlled with lambing results. As a conclusion, the high rate of correct diagnosis were obtained in pregnant and nonpregnant ewes, 84.62 % and 85.72 % respectively. While progesterone values were useful for early diagnosis, oestradiol values were not clear enough to decide the diagnosis.

    Determination of milk and plasma progesterone concentrations for early diagnosing pregnancy in cows

    Summary: The aim of this study was to investigate the pregnancy diagnosis rates by blood and milk serum progesterone assays at the first and second estrous periods after the natural servings in cows and heifers. Fourty Swedish red cows and 10 heifers, between 2- ll years old, were used as materials. Blood and milk samples w ere collecteld from cows in addi tion to blood samples taken from heifers on the days of 21, 38 and 45 days after serving. Blood and milk samples were centrifugated for 20 minutes at the 2000 and 5000 rpm respectively and were kept in at- 20 until the determination of progesterone by radioimmunoassay method.

    In this investigation, 24 Swiss Brown Cows which were clinically healthy and similar age were used. During the two month, the blood samples were taken from dry cows and early, mild an late lactation animals at 15 days intervals. Total thyroxine (TT), total triiodothyronine (TT3), glucose and total cholesterol levels in the blood serum were determined. Daily milk production of the animals in the lactation stages were measured. The mean daily milk production in the early, mild and late lactation cows were found to be 13.94 It, 10.56 It and 8.69 It respectively. It was determined significantly differences among the mean blood serum TT4 (4.19 µg/dl, 4.19 µg/dl, 4.50 µg/dl and 4.64 µg/dl respectively) TT3 (95.67 ng/dl, 106.89 ng/dl, 110, 82 ng/dl and 106.10 ng/dl respectively) glucose (57.94 mg/dl, 58.08 mg/dl, 62.41 mg/dl and 64.35 mg/dl respectively) and total cholesterol (94.34 mg/dl, 93.66 mg/dl, 88.46 mg/dl and 86.11 mg/dl respectively levels of dry animals and cows in the early, mild and late lactation stages. Based on results, it was concluded that lactation was significantly affected the levels of the blood serum TT4, TT3, glucose and total cholesterol depend on daily milk production in the cows.
