| dc.description.abstract | Bu çalışmada, Otojen Staphylococcus aureus aşıları ile süt ineklerinde stafilokokkal mastitislerin insidensinin azaltılması amaçlandı. Klinik ve subklinik mastitisierden izole edilen S. aureus suşları kullanılarak alüminyum hidroksit veya mineral yağ adjuvantlı otojen S. aureus bakterin ve bakterin+toksoid (kombine) aşılar hazırlandı. Otojen S. aureus aşılarının saha şartlarında kurudaki inek ve düvelerin kan ve süt serumlarındaki immúnojenik etkinlikleri (antistafilokokkal, anti - ve B-hemolizin antikorların oluşumu), sütteki somatik hücre sayıları ve meme içi S. aureus enfeksiyonları üzerine etkileri belirlenmeye çalışıldı. Denemeler, 7-8 aylık gebe 15 baş kurudaki Holstein inek ve 12 baş Holstein düve olmak üzere toplam 27 hayvan üzerinde gerçekleştirildi. Otojen S. aureus (bakterin ve kombine) aşıları; meme üstü lenf düğümü bölgesinden deri altı veya sağ uyluk bölgesinden kas içi yolla 2 kez uygulandı. Kontrol grubu hayvanlara göre aşılanan kurudaki inek ve düvelerin kan ve süt serumlarında yüksek titrede antistafilokokkal (P<0.001), anti u-hemolizin (P<0.05) ve anti B-hemolizin (P<0.05) antikorlar belirlendi. Aynı zamanda, aşılı grupların sütlerindeki somatik hücre sayıları kontrol gruplarına göre daha düşük bulundu (P<0.05). Bir laktasyondaki S. aureus mastitisien; aşılı kurudaki ineklerde % 9.3-12.5 oranlarında iken kontrol grubunda % 15.6 oranında tespit edildi. Aşılı duvelerde ise S. aureus mastitisleri % 8.3-12.5 oranlarında bulunurken aşısız düvelerde % 33.3 oranında belirlendi. | en_US |
| dc.description.abstract | The aim of the study was to investigate autogenous Staphylococcus aureus vaccines in control of mastitis in dairy cows. Selected strains of S. aureus isolated from both clinical and subclinical cases were used to prepare autogenous S. aureus vaccines (bacterin and bacterin + toxoid) in aluminium hydroxide or mineral oil adjuvants. Sera and milk samples collected from dry cows and heifers were analysed for immunogenic activity of S. aureus vaccines by detection of antibody titers to staphylocoocal antigens, a- and B-haemolysines, influence on somatic cell counts and intramammary S. aureus incidence. Experimental studies were carried out on a total 27 animals (pregnant for 7-8 months 15 dry cows and 12 heifers). Autogenous S. aureus vaccines were administered to cows by subcutaneous route in the area of supramammary lymph node or intramuscular route to semitendinous muscle. In comparison with control groups, antibody levels to staphyloccal antigens, and to the u- and B- haemolisines were significantly higher (P<0.001, P<0.05 and P<0.05, respectively) in the sera and whey from vaccinated cows. The average somatic cell count in the milk samples of vaccinated cows were also found to be significantly less (P<0.05) than nonvaccinated. cows. The ratio for the S. aureus mastitis in a lactating period, in dry cows vaccinated bacterin and combined vaccines was 12.5 % and 8.3 %, respectively, 15.6 % in nonvaccinated dry cows. For the vaccinated heifers, the ratios were 9.3% 12.5%, while 33 3% in nonvaccinated heifers. | en_US |
