Siyanür ve Tiyosiyanatın Toksikokinetik Üzerine Sodyum Tiyosülfatin Etkilerinin Araştırılması

| dc.description.abstract | This study was made lo delermine the some loxicokinetic parameters of eyanide and thiocyanate in sheep experimentally poisoned with amygdalin In this study, 24 sheep 50 · 60 kg weights were used. Amygdalin and sodium thiosulfale were given and tha eyanide and Ihiocyanate levels in samples were determined Then some toxicokinetic parameters ol lhe eyanide and thiocyanate in the blood samples taken al 0.5, 1,2,4, 8, 24, 48 and 72 th hours were established. The sodium Ihiosullale wera given intravenously in doses 50 and 600 mg!kg and lhen the distribution halllivas ol blood eyanide lavals ware respectively, 0.57±O.05 and 3.24±O.16 hours; the elımination halllives were respectively, 172.93tl1.4 and 77.08tI5.26 hours: mean residence times were respectivety, 250.37±25.2 and 102.1 0i8.12 hours; the duralion of maximal concentration were respectively, 48th hour; the maximal blood levels respectively, 0.177 and 0.186 ,ıg/ml were determıned. Besides. area under the curve values according to doses were respeclively. 26.68±2.18 and 14.20±1.94 Ilg.hour/ml: volume ol distribution at steady state were respectivety, 0.371±O.01 and 4.312±O.77 lJkg: clearance values were respectively, 1.87t1.10 and 42.23t5.64 mVhour.kg were calcutated. The dislribution half·llvas ol blood thiocyanate levels according lo doses were respectivety, 0.44±O.03 and 0.48±O.05 hour; the eliminalion hall·lives were respeclively, 56.17t5.34 and 128.24±8.43 hours; mean residanca times were respectivety, 81.69±4.47 and 152.11±11.6 hours; the duralion ol maximal concentratiorı respectivety, 24 and 21h hours; the maximal blood levels were respeClively, 0.03 and 0.05 Ilg/ml were determined. 8esides, area under the curve values according to doses were respectively, 2.10t0.05 and4.31±O.04llg.hour/mt; volume ol distribution at steady state were respectivety, 1.89±O.03 and 21.278±1.88 lIkg: clearance values were respectivety, 23.75;t6.40 and 139.16t51.7 mVhour.kg were established. | en_US |
| dc.description.abstract | Bu çalışma deneysel olarak amigdalin ile zehirlenen koyunlarda kan siyanür ve tiyosiyanat düzeylerine ait bazı toksikokinetik parametreler üzerine sodyum tiyosülfatın etkilerini araştırmak amacıyla yapıldı. Bu çalışmada, ağırlıkları S()..60 kg arasında olan 24 koyun kullanıldı. Amigdalin ve sodyum tiyosülfatın verilmesinden sonra, 0.5, 1, 2, 4, 8, 24, 48 ve 72. saatlerde alınan kan örneklerindeki siyanür ve tiyosiyanal düzeylerine ait bazı toksikokinetik parameireler belirlendi. Sodyum tiyosülfat 50 ve 600 mW1<g dozlarında damar içi yolla verildi. Oaha sonra kan siyanOr düzeylerinin dağılım dönemi yan ömürlerinin dozlara göre sırasıyla, 0.S7±O.05 ve 3.24±O.16 saat: atılım dönemi yan ömOrlerinin sırasıyla, 172.93t11.4 ve 77.08t15.26 saat; ortalama kalış surelerinin sırasıyla. 2S0.37±2S.2 ve 102.10i8.12 saal; doruk siyanür yoğunluğuna ulaşma süresinin 48.saatler; doruk siyanur düzeylerinin ise sırasıyla, 0.177 ve O.l86llg/ml olduğu ıespit edildi. Bunun yanında siyanür duzeylerine ait eğri altında kalan alan değerleri sırasıyla, 26.68±2.18 ve 14.20±1.94 J1g saaVml; kararlı durumda dağılım hacmi değerlerinin sırasıyla, 0.371±O.01 ve 4.312±O.77 Llkg ve ktirens değerlerinin yine sırasıyla 1.87±1.10 ve 42.23±5.64 mVsaal kg olduğu belirlendi. Kan tiyosiyanat duzeylerine ait dağılım dönemi yan ömürleri dozlara göre sırasıyla O.44±O.03 ve 0.48±O.05 saat: atılım dönemi yarı ömürleri sırasıyla, S6.17t5.34 ve 128.24t8.43 saat; ortalama kalış sureleri sırasıyla, 81.69±4.47 ve 152.11±11.6 saal; doruk siyanur yoğunluğuna ulaşma sureleri sırasıyla, 24 ve 2. saal; doruk siyanür düzeyleri ise sırasıyla, 0.03 ve 0.05 mg olarak belirlendi. Buna ilaveten kan siyanür düzeylerine ait eğri altında kalan alan değerlerinin sırasıyla, 2.1O±Q.05 ve 4.31±O.041l9 saatlml; kararlı durumda dağılım hacmi değerlerinin sırasıyla, 1.89±O.03 ve 21.278±1.88 Llkg ve klirens değerlerinin ise sırasıyla, 23.75±6.40 ve 139. 16±51.7 mVsaal kg 'olduğu tespil edildi. | en_US |
