Öğe Amigdalin ile Zehirlenen Koyunlarda Siyanür ve Tiyosiyanatın Rumen Sıvısı ve Kandaki Bazı Toksikokinetik Parametrelerinin Araştırılması(Selçuk Üniversitesi, 1998) Pirinçci, İbrahim; Tanyıldızı, Sadettin; Kaya, Sezai; Yarsan, Ender; Özaydın, SongülThis investigalion was carried out lo establish some toxicokineıic parameters of the eyanide in rumen liquor and eyanide and ıhiocyanate in blood ol ıhe sheep experimenıally poisoned with amygdalin. In this study , 20 sheep 40 - 50g weights were used. Amygdalin was givcn oraJly in doses ol 0.25, 0.5, 1 and 1.5 m9'k;g. The eyanide and Ihi· ocyanate levels in samples were determined. Then some toxicokinetic paramelers of the eyanide and Ihiocyanate like as the distribution half-life, the elimination half-life, mean residence times and area under Ihe curve (AUC) in rumen liquor and the blood samples laken at 0.5, 1, 2, 4, 8, 24, 48 and 72 Ih hours were eslablished. The distribution half· lifes ol eyanide levels in rumen liquor were respectively, 0.18iO.009, 0.14iO.Ol. 4.66iO.l1 and 4.82tO.17.hours; the elimination half-lifes were respectively, 17.22±O.66, 14.8O±O.43, 9.21iO.16 and 9.66iO.40 hours; AUe values ac· cording lo doses were respeclively, 30.68±O.65, 3O.04iO.24, 38.57iO.22 and 39.29±O.17 mg.hour/ml were observed. Besides mean residence times were respectively. 26.52±1.01, 23.66iO.33, 19.55iO.12 and 19.72±O.3S hours; the duration ol maximal eyanide concentration were respeclively, 2, " 8 and 8th hour; and the maximal eyanide levels ac· cording to were respeclively, 0.983. 1.025, 1.241 and 1.225 ?glml were determined. The distribution half-liles of blood eyanide levels were respectively, O. IStO.OS. 0.42±O.04, 1.4S±2.05 and 9.73±3.74 hours; the elimination hall·lifes were respectively, 59.48±4.40, 55.12±42.66, 70.77±4.10 and 48.05±19.27 hours; and AUe values according lo doses were respectively, ı8.45iO.85. 45.10±21.14, 27.45±2.02 and 2S.76±2.77 Jl9.hour/ml were established. Besides mean residence times ol eyanide levels in blood were respectively, B7.56t5.93, 86.94t54.02, 105.93t7.25 and 75.21±23.10 hours; the duraıion ol maximal concentration 1 hours; and the maximal blood levels according to doses were respeclively, 0.165, 0173, 0.166 and 0.210 mgfml were delermined. The distribution hatf-lifes of blood Ihiocyanate levels were respeclively, 2.05tl.45, O. t6iO.05. O. t7iO.04 and O.29±O.13 hours; the elimination half-liles were respectively, BB.93±2S.08, 73.21±19.44. BO.2BtıOAl and BO.81±15.26 hours; AUe values were respectively, B.60±6.42, 7.47±2.40, 7.14tl.02 and 10.67±3.55 Jlg.hour/ml; and mean residence times according to doses were respectively, 130.56t36.4B, 107.20±27.54. 1 t7.23t14.65 and 117.67±22.01 hours were calculated. Besicles, the duralion ol maximal eoneentralion " 2, 2 and 2 hours; and the maximal blood levels according to doses were respectively, 0.064,0.056, 0.066 and 0.056 mgml were determined.Öğe Pharmacokinetics of Enrofloxacin After Intravenous and Intramuscular Administration in Angora Goats(CANADIAN VET MED ASSOC, 2001) Elmas, Muammer; Traş, Bünyamin; Kaya, Sezai; Baş, Ahmet Levent; Yazar, Enver; Yarsan, EnderPharmacokinetics and bioavailability of enrofloxacin were determined after single intravenous (IV) and intramuscular (IM) administrations of 5 mg/kg body weight (BW) to 5 healthy adult Angora goats. Plasma enrofloxacin concentrations were measured by high performance liquid chromatography. Pharmacokinetics were best described by a 2-compartment open model. The elimination half-life and volume of distribution after IV and IM administrations were similar (t(1/2 beta), 4.0 to 4.7 h and V-d(ss),1.2 to 1.5 L/kg, respectively). Enrofloxacin was rapidly (t(1/2a), 0.25 h) and almost completely absorbed (F, 90%) after IM administration. Mean plasma concentrations of enrofloxacin at 24 h after IV and IM administration (0.07 and 0.09 mug/mL, respectively) were higher than the minimal inhibitory concentration (MIC) values for most pathogens. The cobalt chloride was given intravenously in doses 5 and İS mçVkg and then the distribution half-lives of blood eyanide levels according to doses were respectively, 3.17±O.35 and 4.03±O.65 hours; the elimination half-lives were respectively, 27.62±3.16 and 39.53±5.35 hours; mean residence times were respectively, 44.44±2.76 and 109.29±8.43 hours; Ihe duratlon of maximat coneentration were respectively, 241h hours; the maximal blood levels were respectively, 0.120 and 0128 glml were determined. Besides, area under the curve values according to doses were respectively, 7.09±1.24 and 5.93±O.64 mg. hour/ml; volumes of distribution at steady state were respectively, 0.099±O.OO5 and 0.125iO.Ol l/kg; clearanee values were respeclively, 1.21iO.44 and 2.52±1.21 mVhour. kg were calculated. The distribution half-Ilves of blood thioeyanaıe levels according lo doses were respeclively. 0.30±0.02 and 2.49±1.12 hours; the eliminatian half-lives were respectively, 213.89±11.21 and 96.56±9.35 hours; mean residence times were respectively, 309.09±25.76 and 142.95±15.66 hours; the duralian of maximal concentration were respectively, 2 and ı lh hours; the maximal blood levels were respectively, 0.028 and 0.032 glml were determined. In the study, 200 mice, 40-50 g weights, were used. Sodium eyanide were given intraperiıoneally in doses ol 0,1, 0.2, 0.4 and 0.8 mglkg. Then, some to)(icokinetic parameters ol the eyanide and thiocyanate like as distribution half-lile, elimination half-life, mean existence lime and area under the curve (AUC) in the blood samples drawn at 6,12,30,60,120,240,360 ve 720th minules were determined. The distribution half-lifes ol blood eyanide levels were. respectively, 0.04±O.02, 0.05±O.009. 0.06±O.OO8 and 0.08±O.01th hour; the elimination hall-liles were, respectively, 3.06±O.14, 2.36±O.02, 2.33±O.03 and 2.28±O.06 hours; the duration ol ma)(imat concentraıion was 12 minules and the ma)(imal blood levels, according to doses, respeclively, were 0.153, 0.308, 0.350 and 0.376 ).lg/ml. Besides. mean e)(islence times were, respectively, 4.53±O.20, 3.5O±O.03, 3.46±O.05 and 3.43±O.12 hours and AUC values accordıng to doses were, respectively, 33.50±2.62, 45.40±1.84, 48.91±2.77 and 58.47±2.54 ?ıg.hour/ml were observed. Besides, area under the curve values were according to doses were respectively, 11.28±1.24 and 9±S.62 ?g.hour'ml; volume of distribution at steady slate were respectively, 0.03±O.OOI and 0.2±0.03 Ukg; clearance values were respectively, 0.44±O.12 and 0.16±O.09 ml/hour.kg were calculated.
